The Signature Research Programme in Neuroscience and Behavioural Disorders (“NBD”) is focused on understanding the structure and function of the nervous system, and the neural mechanisms underlying human neurological, psychiatric and ophthalmological disorders.
A Research Assistant position is available in the laboratory of Dr. Jun Nishiyama (https://www.duke-nus.edu.sg/directory/detail/nishiyama-jun). The lab investigates synaptic function and dysfunction in neuropsychiatric disorders including autism, schizophrenia, and Alzheimer’s disease, using cutting-edge molecular biology, genome-editing, optical, transcriptomic and behavioural approaches in mouse models. We are looking for a highly motivated candidate with specialised experience in either (a) bioinformatics analysis or (b) mouse behavioural experiments, and is passionate about the molecular basis of neuropsychiatric disorders.
The selected candidate will perform a variety of research activities within the overall scope of a research project under the supervision of the Principal Investigator or his designate, including but not limited to the following:
(a) Bioinformatics analysis
- Process bulk RNA-seq analysis of mouse brain tissue, from raw reads to differential expression and alternative splicing (e.g. STAR/Salmon, DESeq2, rMATS).
- Process single-cell / single-nucleus RNA-seq analysis: clustering, cell-type annotation and cell-type-specific differential expression (Seurat/Scanpy).
- Integrate with human genetic, imaging and behavioural data using reproducible workflows (e.g. Git, HPC).
(b) Mouse behavioural experiments
- Conduct behavioural test batteries for neuropsychiatric phenotypes (e.g. social interaction, prepulse inhibition, open field, novel object recognition, fear conditioning).
- Perform stereotaxic AAV injection and, where relevant, fiber photometry recording.
- Conduct automated tracking and analysis of behaviour (e.g. EthoVision, DeepLabCut) with blinded, randomised design.
Lab operations and administration
- Assist with general upkeep of laboratory needs such as purchasing of items, tracking of inventory/consumables, maintaining databases, protocols and documentation.
- Prepare reagents and supplies for the team.
- Maintain housekeeping of the laboratory and waste disposal in accordance with guidelines.
Animal work (training will be provided)
- Assist in the mouse colony maintenance such as breeding, plug checks, genotyping and etc.
- Conduct animal-related experiments in compliance with institutional approvals and ethical requirements.
Wet-lab experiments and documentation
- Assist with molecular/cell biology experiments such as PCR, cloning, western blotting, cell/neuronal culture, virus production, immunostaining and etc.
- Conduct accurate monitoring, documenting and reporting of experimental results and/or research findings.
- Perform other related duties incidental to the work described therein.
Job Requirements
- Bachelor’s or Master’s Degree in Biological Science, Neuroscience, Bioinformatics or a related field with minimum 1 year of research experience.
- Have hands-on experience in either (a) RNA-seq and/or single-cell RNA-seq data analysis in R and/or Python (Linux/HPC environment), or (b) independently designing and conducting rodent behavioural experiments.
- Prior experience in stereotaxic surgery, fiber photometry or pose estimation (e.g. DeepLabCut) would be an advantage.
- Willingness to support molecular and cell biology experiments, animal-related research activities, and laboratory administration. Relevant training will be provided.
- Possess strong interpersonal communications skills with excellent written, oral communication and computer skills.
- Have strong attention to detail and a high level of accuracy.
- A strong team player who is able to learn new techniques and work collaboratively in a team environment.
